If you're catholic, the Vatican in depth. If you're not, then a quick visit will be enough. If you want a really good view try climbing to the top of the dome, also in the vatican.
The Colosseum is a must. Next to it are a lot of ancient ruins.
go visit the Fontana di Trevi. Legend says that if you throw a coin inside you'll go to Rome again (at least it worked for my sister).
The Panteon is a nice place to see, and you have a few restaurants (even a mcdonalds) near, some stores to buy souvenirs (even a full roman soldier armor) and is close to the Fontana di trevi.

I would say hit up the Pantheon at night, its beautiful. Plan to eat there but be aware its expensive in a lot of places but very atmospheric. Definitely check out Vatican City, and the Colosseum. I'd recommend picking up a travel guide like one from Frommers for more detailed information.

For Catholics, definitely visit the Vatican - so beauiful, and Michelangelo's Pieta is in the duomo.
Visit the colosseum, circus maximus, palatine hill, forum, pantheon, trevi fountain and imperial fora for Roman historical monuments.
If it isn't too far, Pompeii near Naples is brilliant also.

There really is just so much to do and see. Walk everywhere so you don't miss anything and Rome really is quite small for a city. If you only get to see one thing, I would have to say the Pantheon. I am not a religious person but it was truly awe inspiring. If you are there on a Wednesday, try to get to the Vatican early and get blessed by the Pope! As I said, i'm not religious but it can't hurt can it!

I'd agree with all of the sites listed above. Do toss your coin in (it worked for me, I mostly live here now). If you have more time to do things beyond the regular tourist sites, explore the places real Romans live. Two neighborhoods woth visiting are located on either side of Termini station: San Lorenzo and Piazza Vittorio.
Walk around quickly gentrifying San Lorenzo. See the market, see the stores that sell hand-crafted leather, juggling supplies or world music CDs. Have a coffee or an ice cream with the old-timers at Marani (via dei Volsci 57); have great affordable fish da Franco ar vicoletto (via dei Falisci, 1); or check out any of the very affordable pizzerie or osterie that are filled with students from the nearby university. San Lorenzo is also great for nightlife.
Check out the faded grandeur of Piazza Vittorio, near Termini. Go to MAS, the bizarre dime-store cluttered with everything from overalls to fur coats. Have great Chinese food (the restaurant at via Giolitti 189 is good) or Indian (via Principe Amadeo) or even Roman at Vecchia Roma (via Ferruccio at the corner of via Buonarotti). And have ice cream at Fassi, the oldest ice cream maker in Rome (via Principe Eugenio). The neighborhood is a great mix of working class and upscale (high-end places include the very pricy Agata e Romeo (via Carlo Alberto), the pastry shop Regoli (via dello Statuto) and Pandella, an outstanding bakery (via Merulana).

Colosseo, Foro Romano, Fontana di Trevi, Pantheon, Piazza di Spagna with its famous Spanish steps that lead to Trinità dei Monti, Parco Borghese, Vatican city, Vatican Museum, Castello di Sant' Angelo, Piazza Navona, Piazza Venezia and the Campidoglio, Campo dei Fiori, the famous Via Veneto and if you arrange your trip for Summer, you must go to Trastevere for a drink!

definately get near the colosseo so you can see the roman forum as well. trevi fountain is a must- very different in person! Also yes the Vatican is well worth it catholic or not. I hope you really like history because you will be submerged into it!
